Item An impact of oligoesters of polyatomic alcohol on the content of cytochromes in microsomal fraction of rats’ liver(2016) Бондарева, Алла Владимировна; Жуков, Виктор Иванович; Bondareva, A.; Zhukov, ViktorStudy of mechanisms of pathological processes development is actual one in modern science. Impact of xenobiotics on organism is also important and actual study in modern science. Defined ways of organism adaptation to xenobiotics action were formed during evolution, central role in which presents biochemical mechanisms of their decontamination. Samples of OEF-LP 502 (polyoxypropylene glycol) and 503 (polyoxypropylene triol) were used in this work with defined physical and chemical peculiarities. Experiments were done on sexually mature rats of Wistar with body weight (180-220) gram. Long-lasting impact of OEF-LP 502 and 503 in doses 1/10 and 1/100 LD50 is characterized by destruction of functional condition of rats’ liver microsomes that is defined by decrease of cytochrome Р450 and b5 content. There is also no generally accepted theory that would explain the whole variety of violations and allowed to substantiate the effective complex of treatment and preventive measures for the prevention of reproductive losses and the obstetric complications remote development. The purpose of the study was to study the features of the «proteinase – inhibitor proteinase» system state by detecting the activity of neutrophil elastase and plasma concentrations of proteinase inhibitors α1-antitrypsin and α2-macroglubulin in women with different periods of pregnancy termination. Material and methods. 227 pregnant women who were admitted to the Kharkiv city perinatal center were examined, 190 of them had clinical signs of premature pregnancy termination in the period of gestation of 23–36 weeks. Formation of clinical groups was carried out depending on the term of pregnancy in the form of premature and timely birth. The criteria for including women into groups were the following: young reproductive age, single-pregnancy, absence of gestosis, acute and chronic gynecological and somatic diseases. Detection of preterm labor was carried out in the presence of abdominal pain syndrome and structural changes in the cervix. Conclusions. The disturbance of the dynamic equilibrium between the activity of endogenous proteolytic enzymes and the content of their inhibitors, which is confirmed by the reliable (p ≤ 0,001) increase in the activity of neutrophil elastase, by an average of 46%, together with a 36% increase in plasma α1-antitrypsin plasma levels and a decrease in the concentration of α2-macroglobulin on 30% was observed in women with preterm labor in the period of 23–27 weeks in relation to women with the physiological course of pregnancy, which ended with childbirth without complications in the period of 38–41 weeks. The change in the rates was less pronounce in women with preterm labor in the period of 28–36 weeks in relation to women in the control group: an increase in neutrophil elastase was an average of 25% (p < 0,001) and plasma concentration α1-antitrypsin 21% (p = 0,006) with a decrease concentration of α2-macroglobulin by 13% (p = 0,142). The tension of proteolytic processes was most pronounced in the direction of the proteinase activity prevalence in women with preterm labor in the period of 26–30 weeks, which was confirmed by the probable (p ≤ 0,014) increase in the ratio of the activity of neutrophil elastase to the sum of concentrations of inhibitors of α1-AT and α2-MG on average 32%. The imbalance in the «proteinase – inhibitors of proteinase» system is one of the links in the pathogenesis of non-pregnancy and can contribute in combination with other complications.Item Water-salt and mineral metabolism. Recently, the immunological impair¬ment, as a manifestation of reactions to ecopathogenic condi¬tions, suggests a major pathogenesis role in the development of cardiovascular, neuropsychiatric diseases, as well as diffuse diseases of connective tissue. Objectives - experiment was designed to elucidate the organo¬metric changes in thymus of male rats due to impact of propyl¬ene glycol. 40 WAG matured male rats were divided randomly into two groups. The first group served as a control and constituted 8 animals. The sec¬ond group of 32 rats, 8 rodents in each, were treated via gavage by aqueous solutions of propylene glycol in doze 1/10 LD50 in conversion to 26,38 g/kg during 7, 15, 30, 45 days. All animals were sacrificed on the term defined by experimental design. Thymus specimens were dis¬sected out, and linear dimensions (length, width, height) using digital caliper were measured, along with mass and volume of the thymus. Limits of the thymus morphometric indices’ variability in intact and experimental groups were calculated. The research indicates that exposure to propylene glycol caused marked organometric changes in rats’ thymus. However, more pro¬nounced changes were observed on 7th and 30th days. Were estab¬lished the following limits of variability indices oscillations: IndHL of the experimental group thymus ranged from min=12.57 to max=47.54, the mean value was from 24.67 to 28.02; IndHW of the experimental group thymus ranged from min=11.96 to max=88.73, the mean value was from 36.78 to 41.41; IndT of the experimental group ranged from min = 38.17 to max=141.3, the mean value was from 71.1 to 86.52. The study of the morphometric parameters of the thymus in the experimental group of rats has established a sig¬nificant reduction of all parameters and their deviation from the pa¬rameters of the control group, that shows active reaction of thymus on induced xenobiotic.
