The aim of our research was to investigate erythrocyte membranes of rats with carrageenan-induced gastroenterocolitis using fluorescent probes (ortho-hydroxy derivatives of 2,5-diaryloxazole).Materials and methods. Fluorescence of O1O (2 - (2’-OH-phenyl)-5-phenyl-1,3-oxazole) and PH7 (2-(2’-OH-phenyl)-phenanthro[9,10]-1,3-oxazole) probes in suspensions of erythrocytes of rats with chronic carrageenan-induced gastroenterocolitis was determined and compared to the control group consisted of intact animals.Results. It has been found that the hydration of erythrocyte membranes increases in rats with carrageenan-induced gastroenterocolitis. The disease is associated with the changes in the polar regions of the membranes, while no changes in the hydrophobic areas of the erythrocyte membranes are observed. Conclusion. The detected changes allow us to make a conclusion that the fluidity of the erythrocyte membranes of the rats with chronic carrageenan-induced gastroenterocolitis increases.Item About using modern technologies aided monitoring of glycemic control in the type i diabetes in Ukraine(Salzburg, Austria, 2016) Смирнов, Иван Иванович; Новикова, Ирина Владимировна; Высоцкая, Елена Владимировна; Порван, Андрей Павлович; Масалитин, Евгений Сергеевич; Smirnov, I.; Novikova, I.; Visotskaya, E.; Porvan, A.; Masalitin E.The relevance of investigation due to the constant increase in the number of people suffering from type I diabetes that is diagnosed advantageously amongst the younger population. The article discusses the existing approaches of intensified insulin therapy using individual automatic dispensers for basal-bolus principle. Analysis of the aided monitoring of blood glucose levels has been carried out and a weakness of existing of calculators of boluses has been determined. The operation of such of devices can lead to an inadequate increase of total insulin dose. The solution of this problem is possible when improving existing mathematical models based on base relations, who are used in modern calculators of boluses, and development of intuitive management programs an automated blood glucose control levels. Compounds of imidazoline are compounds of xenobiotics organism components of which according to physical and chemical peculiarities and molecules structure properties contain group of cationic surface active substances. Compounds of imidazoline are characterized by significant synthesis volume, wide use in different fields of economic and intake in water sources and possible impact on population health. Previous investigations determined long-lasting rats’ toxification by compounds of imidazoline with alkyl radicals S7-9 and S9-15 in doses 1/10 and 1/100 DL50 initiates the development of oxidative stress with the destruction of protein and lipid molecule. Impact of xenobiotics on organism is also important and actual study in modern science. Defined ways of organism adaptation to xenobiotics action were formed during evolution, central role in which presents biochemical mechanisms of their decontamination. Samples of OEF-LP 502 (polyoxypropylene glycol) and 503 (polyoxypropylene triol) were used in this work with defined physical and chemical peculiarities. Experiments were done on sexually mature rats of Wistar with body weight (180-220) gram. Long-lasting impact of OEF-LP 502 and 503 in doses 1/10 and 1/100 LD50 is characterized by destruction of functional condition of rats’ liver microsomes that is defined by decrease of cytochrome Р450 and b5 content. Increase of cytochrome level on the 15th day of substances impact is adaptive reaction of organism to xenobiotics intake, which is connected with decontamination process, but on the other hand with generation of active forms of oxygen and products of lipid peroxidation.Item Anticonvulsant activity of Fumaria schleicheri dry extract and sodium valproate: Role of neurotrophin and cytokine pathways(2020-05) Tsyvunin, Vadim; Shtrygol, Sergiy; Gorbach, Tetiana; Shtrygol, DianaIntroduction: Cytokine and neurotrophin pathways can be potential targets for principally new anticonvulsant drugs which in perspective will be probably able to solve the problem of pharmacological resistance for people sick with epilepsy and to avoid polypragmasy which considerably increases the risk of developing side effects. Objective: The role of neurotrophin and cytokine link in anticonvulsant activity of Fumaria schleicheri Soy.-Will. dry extract and sodium valproate was assessed by the change in the cerebral level of NGF and IL-1β, TNF-α, IL-4. Materials and Methods: A total of 51 mice were used. The animals were divided into 4 groups: intact control, pentylenetetrazole, pentylenetetrazole + F. schleicheri dry extract, pentylenetetrazole + sodium valproate. F. schleicheri dry extract (100 mg/kg) and sodium valproate (300 mg/kg) were administered into the stomach once a day for 3 days. Pentylenetetrazole (80 mg/kg) was administered subcutaneously on the 3rd day for 30 minutes after the introduction of the tested drugs. After 18 hours the content of cerebral NGF and neurotropic cytokines was determined in the surviving animals. Results: Both F. schleicheri dry extract and sodium valproate provide an expressed anticonvulsant activity, resulting in a significant increase in the latency of the first seizure and reducing the number of clonic-tonic convulsions in 1 mouse, % of mice with tonic seizures and the duration of the convulsive period in the group. Pentylenetetrazole causes considerable changes in both neurotrophin NGF, and cytokines with the proved neurotropic properties – IL-1β, TNF-α и IL-4. F. schleicheri dry extract as sodium valproate, decreased pentylenetetrazole-associated NGF overexpression, normalized the content of IL-1β, TNF-α and increased the level of IL-4 compared not only with the group of animals that were getting pentylenetetrazole, but also compared with intact control. The significant correlation relationship was marked only in the seizure control between TNF-α and IL-4. Conclusion: Thus, the experimental evidence supports a role of NGF and IL-1β, TNF-α and IL-4 on anticonvulsant activity of F. schleicheri dry extract and sodium valproate and suggests potential targets for perspective anticonvulsant drugs.Item Antimicrobial and antioxidant activity of anthocyanin complexes of some berries' species of Ukraine(Чорноморський національний університет ім. The main goal of the current study was to investigate oxidative damage of proteins and lipids in patients with the severe combined thoracic trauma on the first days after injury. The second aim was to evaluate the possibility of outcome prediction through oxidative stress markers determination. Concentrations of malonic dialdehyde and proteins carbonyl groups were determined according to spectrophotometric methods. As there were no differences between survival and nonsurvival groups of patients for oxidative stress markers, cluster analysis was performed for stratification of patients’ population by both oxidative stress markers simultaneously. It was found that oxidative stress develops from 1-2-nd day after the severe combined thoracic trauma and generally its level can be reliably estimated through determination of relative concentrations of both malonic dialdehyde and proteins carbonyl groups. The degree of oxidative stress is proportional to the severity of injury and patients’ state on admission (traumatic shock) and does not depends on patients’ age and concomitant alcohol exposure. Also its progression is not linear as the result of severe drop of the oxidative stress markers` concentration in premorbid phase of wound dystrophy. However, behavioral and cognitive impairment in rats at the different stages of vascular model of dementia of Alzheimer's type developed by us has not been investigated. Subjects and methods. The experiment was performed on 32 male WAG rats weighing 180–250 g which were divided into 4 groups. Rats from group 1 and 3 were injected aqueous solution of sodium nitrite at a dose of 50 mg/kg of body mass intraperitoneally during 14 and 28 days respectively. Groups 2 and 4 received 500,000 mesenchymal stem cells in suspension intravenously against a background of experimental nitrite-induced AD. To estimate the behavioral reactions and cognitive functions the Open Field Test (OFT) and Passive Avoidance test (PAT) were used. Results. In all experimental groups in most cases significant decrease in vertical and horizontal activity (p <0.05) and an increase in the number of defecation in the OFT were found. Rats from group 3 had the drop in locomotor, research and orientation activity. In the OFT and PAT in groups 2, 4 improvement in research activity and significant cognitive functions recovery was observed (p=0.012) after stem cell correction. Conclusion. Progression of the protective inhibition and cognitive impairment was found during the experiment. Stem cell administration had positive effects on brain function recovery.Item Behavioural Reactions of Random-Bred Mice under the Influence of Hypouricemia and Aegopodium podagraria L. Still, their efficacy was not determined in combined use with therapeutic doses of allopurinol. This study addressed the changes of uric acid metabolism and CNS in mice undergoing hyperuricemia correction with allopurinol combined with A. podagraria extract or tincture. Study Design: The mice were randomly distributed to five groups: Group I: intact control; Group II: control for manipulations and hypouricemia (allopurinol, 2.5 mg/kg); Group III: potassium oxonate (PO), 250 mg/kg + allopurinol, 10 mg/kg; Group IV: PO, 250 mg/kg + allopurinol, 10 mg/kg + extract, 1 g/kg; Group V: PO, 250 mg/kg + allopurinol, 10 mg/kg + tincture, 1 ml/kg. Place and Duration of Study: Central Scientific-Research Laboratory, National University of Pharmacy, Kharkiv, Department of Biochemistry, Kharkiv National Medical University, June 2017 – September 2017. Methodology: Beginning from the 15th day psychopharmacological tests were carried out. At day 21, xanthine oxidase activity in the liver and kidney, uricase activity in the liver, uric acid level in blood and brain, GABA, serotonin, aspartic and glutamic acids concentrations in the brain were determined. Results: The extract as well as the tincture did not counteract the influence of allopurinol on xanthine oxidase, liver uric acid level was decreased, and uricemia slightly elevated (especially by the tincture) allowing to suggest the changes of uric acid transport. Such changes were also possible in brain resulting in the increased uric acid level in animals receiving combinations with A. podagraria or allopurinol per se. In these groups, GABA brain content was reduced, while aspartic and glutamic acids content was increased. The extract and especially the tincture decreased brain serotonin level (which was elevated by PO and allopurinol). Allopurinol per se and its combinations with A. podagraria preparations mildly reduced locomotor activity. Allopurinol and PO increased the duration of stay in the open arms of the elevated plus maze that was eliminated by the tincture, which also normalized the number of mice immediately visiting the open arm. The extract and the tincture decreased depressivity level in the tail suspension test. The tincture restored physical endurance in the weight-loading forced swimming test. Conclusion: The results substantiate the combined use of allopurinol and A. podagraria preparations, which do not counteract the main effect of allopurinol and do not cause the negative changes of the CNS (no unfavourable shifts in locomotion and anxiety are induced, while depressivity and physical endurance are partially improved). Further studies are needed to elucidate the mechanisms of A. podagraria active components interaction with allopurinol within the brain.Item Biochemistry(KNMU, 2012) Popova, Liudmyla; Polikarpova, AnnaThe manual summarizes current knowledge relevant to the static, dynamic, and functional biochemistry.
