Item Antimicrobial Activity Derivatives 2H-pirano[2,3-c]piridines against Pathogens of Intestinal Yersiniosis(Journal of Pharmacy and Nutrition Sciences, 2021) Ivannik, V.; Torianyk, I.; Moiseienko, T.; Skliar, A.; Yeromenko, R.; Hnatiuk, V.; Podrigalo, L.; Nazaryan, R.; Mikhailenko, Nina; Gargin, V.Background: An important aspect in the treatment of patients with intestinal yersiniosis is the administration of effective antibiotic therapy. Performed research aimed to determine the spectrum and level of antimicrobial activity of 2H-pyrano[2,3-c]pyridine derivatives on the museum and clinical strains of gram-negative microorganisms Yersinia enterocolitica. Methodology: The object of the study was 28 synthetic derivatives of 2H-pyrano[2,3-c]pyridine. The compounds were studied according to their chemical structure. Weused the method of serial dilutions in Muller-Hinton liquid nutrient medium with a museum’s and clinical strains of Y.enterocolitica. Results: Studies indicate the promise of further study of the properties of 2Hpyrono[2,3-c]pyridine to create an effective antimicrobial medicine. According to theresults of studies on action of antimicrobial compounds synthesized on the basis of 2H-pyrano[2,3-с]pyridine derivatives, it was found that the MIC of compounds for all Y. enterocolitica strains was 100.0 μg/ml. The MBCC of most cultures of Yersinia (72.3 %) was 200.0 μg/ml. Compound 2{3} had a pronounced antiyersiniotic activity, the inhibitory effect of which was manifested at a concentration of 25.0 μg/ml. Retarding the growth of most Yersinia strains (95.3%) with a MIC of 50.0 μg/ml, the MIC of compounds ranged from 50.0 to 200.0 μg/ml. After statistical data processing, pyridine derivatives (compounds 2{3} and 3{5}) were identified, possessing an effective bacteriostatic and bactericidal effect on Y. enterocolitica strains. Conclusions: The results of the research showed a high antimicrobial activity of 2Hpyrano[2,3-c]pyridine derivatives. Simultaneously microcirculatory disturbance is important link of pathogenesis in many pathological conditions in oral cavity with inflammatory consequences. The aim of this study was to determine importance of microcirculatory disturbance of oral mucosa in modeling of rheumatoid arthritis. Experimental investigation has been performed with modeling RA on laboratory white male mice according to described before method. Investigated groups were formed according to severity manifestation as ankle changes using digital calipers measuring. The specimens of soft tissues of the oral cavity were stained with hematoxylin and eosin, according to van Gieson, according to Rego after the routine proceeding. Morphometric studies were performed with estimation of volumes of specific vascular density in microcirculatory bed, density of connective tissue in lamina propria and area of tissue with ischemia. It was detected that disturbance of oral mucosae microvasculature is formed in rheumatoid arthritis with strong correlation relationship between specific densities of microcirculatory bed vessels and rheumatoid arthritis severity (r=0.74). Development of ischemic area indicates strong correlation relationship between ischemic area and rheumatoid arthritis severity also (r=0.72) and it could be connected with changes in microvasculature (r=0.82). The interrelation of disorders in periodontal tissues with metabolic changes caused, in particular, by a violation of microcirculation, observed namely in smoking but the fact that the effect of smoking on the condition of periodontal tissues at a young age has been studied less than other risk factors deserves special attention. The aim of our study was to experimentally assess the effect of smoking on the morphofunctional state of periodontal tissues in young animals. Materials and methods. Simulation of smoking has been performed with ten-week-old WAG rats using the Boyarchuck chamber. Morphometric studies were performed in the gingival zone which was chosen for morphological interpretation of volumes of specific vascular density in microcirculatory bed, specific density of connective tissue in lamina propria and specific area of tissue with ischemia. The obtained digital data were processed by the methods of variation statistics. Results. Simulation of smoking for four weeks in experimental animals leads to pathological changes in the morphofunctional state of the periodontium with reducing vascular density from 19,44±1,97% to 11,03±1,38 %, increasing area of connective tissue from 18,33±2,71% to 26,49±1,24%, spreading area of tissue with ischemia from 1,14±0,70% to 6,35±1,67%. Conclusions. The aim of the study was to determine the ecological characteristics of the oral microbiota and some antimicrobial factors of saliva in children with mucoviscidosis. Materials and methods. The study involved an assessment of oral microbiota as complex ecological system that protects the human body from colonization by pathogenic flora in children with cystic fibrosis. Bacteriological studies have been performed on clinical material from 30 children with mucoviscidosis diagnosed with dental and periodontal diseases. Results. In the microbiological study of plaque microbiota, 70 strains of opportunistic pathogens were isolated in patients with mucoviscidosis. The most significant were alpha-hemolytic Streptococci (40%). The proportion of bacteria of Neisseria genus in patients with cystic fibrosis was lower and amounted to 24.3%. C. albicans fungi were isolated in comparable values (18.5%), S. aureus (8.5%), as well as gram-negative strains of E. aerogenes (4.3%) and E. coli (4.3%) significantly dominated. The results indicate that opportunistic pathogens S. aureus, E. aerogenes and E. coli partially replaced the representatives of the normal oral microbiota alpha-hemolytic streptococci and non-pathogenic species of Neisseria genus in patients with mucoviscidosis. Conclusions. Microbiota of plaque in children with mucoviscidosis is characterized by an expansion of the spectrum of opportunistic pathogens due to Staphylococcus aureus, enterobacteria and C. albicans fungi, which indicates a violation of the microbiocenosis due to reduced mucosal immunity. The aim of this study was to determine the effects of ozone on the morphofunctional peculiarities of the soft tissues in modeling chronic recurrent aphthous stomatitis. We performed experimental investigation for study of the morpho-functional state of tissues of the oral mucosa in CRAS with using of previously proposed and widely used modeling scheme with ovalbumin and aluminum hydroxide. Two groups of animals were formed (Dutch rabbits, males, aging three-month, weighting 2-2.4 kg). Group of 8 animals with obtained mucosal changes was our comparison group. Other group of 8 animals with obtained mucosal changes was treated by ozone therapy. Histological investigation has been performed. Microscopical examination of tissue had shown that ozone therapy reduces inflammation and edema and is useful in wound healing in soft tissue as disappearance of necrobiotic processes, epithelialization of aphthous defect, growth of akantotic bands, pronounced reducing of inflammatory cells and changing of cellular ratio (with of neutrophils part from 38.30±2.46% to 6.34±0.63%, eosinophils from 5.49±0.23% to 2.87±0.05%), restoration of the cellular layers of the epithelium, moderately pronounced sclerosis of the papillary layer of the lamina propria. Described results allow to conclude that correction of tissual changes in chronic recurrent aphthous stomatitis could be obtained with ozone therapy using.Item Influence of the complex treatment of chronic generalized catarrhal gingivitis on microbiological and immunological parameters of oral cavity of children with cystic fibrosis(2017-10) Nazaryan, R.; Tkachenko, M.; Kovalenko, NatalyaЗ метою попередження розвитку інфекції при хронічному генералізованому катаральному гінгівіті у дітей, хворих на муковісцидоз, запропоновано лікувально-профілактичний комплекс, який поєднує застосування гелю, що містить хлоргексидину біглюконат, амінофторид та бетаїн; суміші 12,5% розчину кислоти акридоноцтової і 0,9% розчину натрію хлориду для ротових ванночок; ультразвукової та електротерапії із засобом, що містить 0,01% розчину мірамістину. У результаті проведених лікувально-профілактичнх заходів відбувалося відновлення нормальної мікрофлори зубного нальоту за рахунок α-гемолітичних стрептококів і нейсерій, що спричинило зменшення у 1,6 рази формування асоціацій мікроорганізмів і витіснення S. aureus та зниження частки C. albicans з 18,6 % до 7,4 %. Крім того, спостерігалося збільшення продукції лізоциму та секреторного імуноглобуліну. Проте через 3 місяці знову відбулася колонізація дослідженого біотопу золотистим стафілококом, а кількість грибів C. albicans зросла до 14,7 %.Item Interrelation of the gene CDKN1A (RS 1801270) polymorphic state аnd level of development of caries in children with Down syndrome(2018) Nazaryan, R.; Iskorostenskaya, O.; Gorenskaya, O.; Volkova, N.; Gargin, VitaliyStudy is devoted for rs1801270 polymorphism of CDKN1A gene due to the level of intensity of caries development in children with Down syndrome. The full development of oral cavity organs is ensured through a complex of genes whose protein products control different stages of this difficult process. Changes in gene structure, as well as disturbance of their expression may lead to abnormalities of development of teeth and periodontal tissues. Analysis of the polymorphism of CDKN1A gene Ser31Arg in children living in Kharkiv region has shown that the frequency of genotypes and alleles corresponds to the theoretically expected distribution by Hardy-Weinberg in the core (χ2=0.12) and control (χ2=1.29) groups. There is no significant differences in the frequency of alleles of CDKN1A gene in the group of healthy children and children with Down syndrome (p=0.978). To simulate the atopic process, young experimental animals (threemonth- old male rabbits) were sensitized by intraperitoneal injection of ovalbumin and aluminum hydroxide during the first 3 days of the experiment. Twice lower dose of ovalbumin was instilled intranasally under local anesthesia five days later (Day 8) with repeated intranasal administration of ovalbumin on the 16th, 17th, 20th and 21st day of the experiment. Obtained specimens of oral cavity were examined histologically, and immunohistochemical study was performed to determine the immunoreactivity of eNOs, iNOs, CD23, CD20. Histological investigation of obtained microslides detected that atopic modeling process is implemented by a complex of pathological changes of oral mucosa with the presence of intraepithelial lymphocytes, eosinophils, focal erosive lesions, signs of proliferation of the basal cell layer, moderately expressed papillomatosis. Such histological picture can be interpreted as the development of inflammatory, degenerative, dyscirculatory process. It was found that such changes are accompanied by disturbance of nitric oxide synthase metabolism, characterized by increased activity of inducible nitric oxide synthase more than twice and endothelial synthase in the extravascular space. Inflammatory infiltrate in atopic process is presented by B-lymphocytes, activated macrophages, eosinophils both in the lamina propria and epithelium that is indicated by a sharp increase in the immunoreactivity of CD23 and CD20. Accumulation of these cells has strong correlation dependence with nitric oxide synthase. The most pronounced correlation has been detected between CD23 and CD20 (r=0.89), iNOs and CD23 (r=0.85), iNOs and CD20 (r=0.87) while comparing the results of immunohistochemical study with eNOs, iNOs, CD23, CD20.
